Flag Peptide, Flag peptide tag, A fusion tag known as DYKDDDDK, composed of eight amino acids (Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys) and including an enterokinase-cleavage site, was specifically designed for use in immunoaffinity chromatography. This tag enables elution under non-denaturing conditions. Several antibodies targeting this peptide have been developed. One such antibody, referred to as M1, binds the peptide in the presence of divalent metal cations, preferably Ca²⁺. Elution can be achieved using chelating agents or alternatively through competitive elution with an excess of free DYKDDDDK peptide.
